Applications : This tricyclic benzopyrimidodiazepinone was identified as a nanomolar‑potency ACK1 inhibitor, demonstrating strong biochemical activity against the kinase. However, despite its promising potency, the scaffold suffered from poor pharmacokinetic properties, which ultimately prevented further development and highlighted the need for more drug‑like chemotypes in ACK1 inhibitor optimization.
