Applications : Amitriptyline exerts antidepressant activity through potent inhibition of SERT and NET, with Ki values of 3.45 nM and 13.3 nM, respectively, leading to increased synaptic 5‑HT and norepinephrine. It also activates α₂A and TrkA/TrkB receptors, producing analgesic and neuroprotective effects. Amitriptyline interacts with a broad receptor spectrum—muscarinic M₁–M₅, H₁, and α₁‑adrenergic receptors—and binds DAT at micromolar affinity. Its ion‑channel blockade, including sodium channels and hERG (IC₅₀ = 4.78 µM), underlies cardiotoxic potential.
