Applications : Amitriptyline hydrochloride exerts antidepressant effects through strong inhibition of SERT and NET, with Ki values of 3.45 nM and 13.3 nM, respectively, leading to increased synaptic monoamines. It also activates α₂A and TrkA/TrkB receptors, contributing to analgesic and neuroprotective properties. The compound interacts with a broad receptor spectrum—muscarinic M₁–M₅, H₁, and α₁‑adrenergic receptors—and binds DAT at micromolar affinity. Its ability to block sodium channels and the hERG potassium channel (IC₅₀= 4.78 µM) is associated with cardiotoxicity. Amitriptyline hydrochloride further exhibits anti‑inflammatory, anti‑angiogenic, and anti‑fibrotic effects.
