Applications : B9 is a potent and selective LXRβ agonist (IC₅₀ = 0.078–0.36 μM across analogs) derived from a spiro[pyrrolidine‑3,3′‑oxindole] scaffold. It strongly inhibits RANKL‑induced osteoclast differentiation and bone resorption in vitro and in vivo. B9 activates LXRβ to promote intracellular cholesterol efflux and reduce extracellular cholesterol uptake, thereby suppressing osteoclastogenesis. These properties position B9 as a promising LXRβ‑targeted agent for osteoporosis therapy development.
