Applications : Divarasib is an orally active KRAS^G12C inhibitor with sub‑0.01 µM potency. By covalently engaging Cys12 in GDP‑bound KRAS^G12C and occupying the switch II pocket, it blocks GTP loading and SOS‑mediated reactivation, effectively shutting down KRAS‑dependent signaling. Divarasib induces significant tumor shrinkage and growth inhibition in KRAS^G12C‑positive cancer models, including lung, colorectal, and pancreatic tumors.
