Applications : Divarasib adipate is an orally active KRAS^G12C inhibitor with sub‑0.01 µM potency. By covalently engaging Cys12 in GDP‑bound KRAS^G12C and occupying the switch II pocket, it blocks GTP binding and SOS‑mediated reactivation, effectively shutting down KRAS‑dependent signaling. The adipate form induces significant tumor shrinkage and growth inhibition in KRAS^G12C‑positive cancer models, including lung, colorectal, and pancreatic tumors.
