Applications : This is a potent fourth‑generation allosteric EGFR inhibitor designed to overcome EGFR^L858R/C797S resistance to third‑generation TKIs. It shows improved potency over earlier allosteric inhibitors and is the first to induce robust tumor regression in an EGFR^L858R/C797S model. Itis also active in H1975 (EGFR^L858R/T790M) xenografts and demonstrates superior efficacy in combination with osimertinib. These properties support as a promising agent for EGFR‑driven NSCLC, both as a single agent and in combination therapy.
