Applications : This is a novel and selective FXIa inhibitor that engages additional interactions within the S2 subsite of FXIa. It demonstrates strong inhibitory potency against FXIa and high selectivity over PKal as well as several other serine proteases. Functionally, it produces significant anticoagulant activity through the intrinsic pathway while sparing the extrinsic pathway. In vivo, the compound shows marked antithrombotic efficacy without increasing bleeding risk or causing observable toxicity in mice, indicating its potential as a promising candidate for further development.
