Applications : Genz‑667348 is a potent, selective Plasmodium DHODH inhibitor, showing low‑nanomolar potency against P. falciparum, P. vivax, and P. berghei DHODH while sparing human DHODH. It demonstrates strong in vitro activity against both 3D7 and Dd2 malaria strains, good oral exposure and tolerability in mice, and ED₅₀ values of 13–21 mg/kg/day in the P. berghei 4‑day murine model. Notably, 100 mg/kg/day oral dosing achieved sterile cure, highlighting Genz‑667348 as a promising anti‑malarial lead for further development.
