Applications : JN-122 is a highly potent spiroindoline‑based MDM2 inhibitor discovered through structure‑guided design. It blocks the MDM2/p53 interaction and also shows moderate inhibition of MDM4/p53, enabling dual relief of p53 suppression. In wild‑type p53 cancer cell lines, JN-122 robustly activates p53 signaling, induces cell‑cycle arrest, and triggers apoptosis, while also promoting MDM4 degradation. The compound demonstrates good PK properties and delivers strong antitumor efficacy in a systemic MOLM‑13 xenograft model. This spiro‑indoline scaffold represents a promising platform for developing next‑generation MDM2 inhibitors targeting cancers with intact p53.
