Applications : MRTX-1133 engages the switch II pocket of KRAS^G12D through a noncovalent alkyne‑based scaffold and binds with extraordinary affinity (Kd ≈ 0.2 pM). It inhibits SOS1‑driven nucleotide exchange and prevents assembly of the KRAS^G12D/GTP/RAF1 signaling complex, resulting in potent suppression of downstream oncogenic pathways. MRTX-1133 selectively targets KRAS^G12D‑mutant cells and shows strong cellular and in‑vivo antitumor activity.
