Applications : This is a brain‑penetrant catalytic mTOR inhibitor emerging from SAR optimization to eliminate the genotoxicity risk observed in the earlier lead (compound 1). It effectively suppresses aberrant mTOR hyperactivity and improves survival in neuronal Tsc1‑knockout mouse models, demonstrating strong CNS‑target engagement. Although oral exposure in higher species is limited, compound 9 remains a valuable CNS tool molecule for probing mTOR‑driven neurobiology and disease mechanisms.
