Applications : This nicotinamide‑based NaV1.8 inhibitor bearing a bicyclic aromatic fragment shows high potency (IC₅₀ ≈ 50 nM in human NaV1.8‑expressing HEK293 cells), retains strong inhibition in DRG neurons, and exhibits excellent isoform selectivity (>200‑fold vs NaV1.1, NaV1.5, NaV1.7), reducing CNS and cardiac liabilities. In vivo, it delivers clear analgesic efficacy in a post‑surgical mouse model, supporting its promise as a non‑addictive, peripherally acting NaV1.8‑targeted analgesic lead.
