Applications : This is a potent HIV‑1 NNRTI based on a 2,4,5‑trisubstituted pyrimidine scaffold designed to exploit tolerant regions of the NNRTI binding pocket. It shows excellent antiviral potency against wild‑type and multiple NNRTI‑resistant HIV‑1 strains (EC₅₀ = 3.14–22.1 nM), outperforming etravirine. It exhibits favorable PK (T_1/2 = 1.33 h, F = 31.8%) and a strong safety profile (LD₅₀ > 2000 mg/kg). Molecular modeling indicates improved resistance tolerance, positioning as a promising next‑generation NNRTI candidate.
