Applications : This is a bicyclic HIV‑1 NNRTI derived from etravirine/rilpivirine scaffolds, incorporating a modified heterobicyclic core and an acrylonitrile substituent. It shows single‑digit nanomolar potency against WT HIV‑1 (EC₅₀ = 3.0 nM) and retains low‑nanomolar activity against HXB2 and key NNRTI‑resistant strains (K103N, Y181C; fold change 1.2–6.7×).
