Applications : This is a cyclopropyl‑linked small‑molecule PD‑1/PD‑L1 inhibitor showing potent activity against the PD‑1/PD‑L1 interaction (IC₅₀ = 29 nM) and strong PD‑L1 binding (K_D= 1.554 × 10⁻¹ µM). It reduces tumor‑cell survival in H460/Jurkat co‑culture assays, has favorable metabolic stability, and demonstrates good PK with ~22% oral bioavailability. In LLC1 lung carcinoma models, it shows robust antitumor efficacy without observable toxicity, suppressing tumor growth by activating the immune microenvironment, making it a promising PD‑1/PD‑L1 inhibitor lead for further development.
