Applications : This novel PDE1 inhibitor shows potent PDE1C activity (IC₅₀ = 10 nM) with high selectivity across other PDE isoforms except PDE4 and only weak calcium‑channel antagonism. It demonstrates strong antifibrotic efficacy in a bleomycin‑induced pulmonary fibrosis rat model and prevents TGF‑β1–driven myofibroblast differentiation. Elevated PDE1B/C expression in fibrotic foci and increased lung cAMP/cGMP levels support a cAMP/cGMP‑mediated mechanism. Overall, this compound represents a promising PDE1‑targeted antifibrotic lead.
