Applications : This a potent and selective RORγt agonist derived from biaryl amide optimization using a “functionality‑switching” strategy starting from the inverse agonist GSK-805. It shows strong RORγt activation, markedly improved metabolic stability, and excellent in vivo PK. In preclinical tumor models (B16F10 melanoma and LLC lung adenocarcinoma), compound 14 delivers robust antitumor efficacy, highlighting it as a promising RORγt agonist lead for cancer immunotherapy.
