Applications : This is a potent and selective tricyclic RORγt agonist developed through structure‑based optimization of an initial benzyloxy aryl hit. It exhibits favorable oral pharmacokinetics and a clear dose‑dependent pharmacodynamic response in the MC38 mouse tumor model. Despite these strengths, no detectable antitumor efficacy was observed at the evaluated doses, indicating that further optimization or alternative models may be required for therapeutic advancement.
