Applications : This is a potent guanidine‑based SHP2 allosteric inhibitor (IC₅₀ = 17.7 nM) that effectively suppresses proliferation, migration, and invasion of MIA PaCa‑2 pancreatic cancer cells. It demonstrates strong in vivo PK (AUC_po = 4320 nM·h; F = 66.3%) and delivers significant antitumor efficacy in the MIA PaCa‑2 xenograft model. This scaffold offers a promising strategy for targeting KRAS‑mutant cancers, while potentially mitigating safety risks associated with the alkyl‑amine motifs found in current SHP2 allosteric inhibitors.
