Applications : This is a novel TAAR1 agonist engineered to activate both TAAR1‑Gs and TAAR1‑Gq pathways, unlike SEP‑363856, which engages only the Gs pathway. It markedly improves MK‑801‑induced schizophrenia‑like cognitive deficits in rodents without causing catalepsy, demonstrating a differentiated and favorable pharmacology. The HCl salt shows good PK (t_1/2= 2.31 h; F = 39%) and a solid safety profile, supporting its potential as a next‑generation TAAR1‑targeted antipsychotic aimed at improving cognitive outcomes in schizophrenia.
