Applications : TAS‑121 is an orally active, covalent third‑generation EGFR inhibitor with potent activity against sensitizing and T790M resistance mutations. It inhibits EGFR^L858R, Ex19del, L858R/T790M, and Ex19del/T790M at sub‑nanomolar to low‑nanomolar concentrations and shows moderate activity toward EGFR^WT. TAS‑121 additionally inhibits HER2 and HER4. By blocking EGFR phosphorylation and downstream signaling, it suppresses proliferation and induces apoptosis, producing significant antitumor effects in SW48 and NCI‑H1975 xenograft models.
