Applications : Thiorphan is a selective neprilysin (NEP) inhibitor with an IC50 of 6.9 nM that competitively blocks NEP activity, preventing neuropeptide degradation such as substance P (SP) and neurokinin A (NKA). In neonatal brain injury research, Thiorphan raises SP and NKA levels, activates NK1/NK2 receptors and related pathways, inhibits overactive NMDA receptors, and protects neocortical neurons from excitotoxicity. It may also reduce NEP-driven bronchoconstriction, making it relevant for respiratory disease studies.
