Applications : This is a new class of potent, highly selective TNIK inhibitors was identified, with SAR studies yielding a lead showing strong TNIK inhibition (IC₅₀= 0.026 ± 0.008 μM) and excellent selectivity across 406 kinases. It effectively suppressed CRC cell proliferation and migration in vitro and demonstrated notable antitumor activity in the HCT116 xenograft model, along with favorable pharmacokinetic properties. This compound represents a promising TNIK‑targeted lead for further drug development.
