Applications : This Evodiamine‑derived polycyclic heterocycle was developed as a Top1 inhibitor for TNBC, with the lead compound showing potent activity (IC₅₀ = 0.36 µM), driving Top1–DNA covalent complex formation and ROS‑mediated DNA damage, and delivering significant antitumor efficacy in a TNBC patient‑derived xenograft model.
