Applications : This is a macrocyclic type II TRK inhibitor with strong potency against wild‑type TRK and key resistance mutations (SF, GK, xDFG). It inhibits TRKA/C, G595R, G667C, and F589L in the low‑nanomolar range and shows high kinome selectivity. It suppresses proliferation of multiple Ba/F3 TRK mutants and demonstrates robust antitumor efficacy in G595R and G667C xenograft models, making it a promising pan‑TRK lead for overcoming clinical resistance.
