Applications : This 1,2,4‑triazolo[1,5‑a]pyrimidine (TPD) derivative is part of a new MT‑stabilizing series optimized for Alzheimer’s disease and neurodegeneration. Substituent‑dependent phenotypes indicate engagement of two tubulin binding sites (seventh and vinca). Systematic SAR and computational studies identified an MT‑normalizing TPD lead with brain penetration, oral bioavailability, and demonstrable brain PD activity, representing a promising microtubule‑targeted neurotherapeutic candidate.
