Applications : YKL‑5‑124 TFA irreversibly and covalently inhibits CDK7 with IC₅₀ values of 53.5 nM (CDK7) and 9.7 nM (CDK7/Mat1/CycH). It is >100‑fold more selective for CDK7 than for CDK9 or CDK2 and remains inactive against CDK12 and CDK13. Biologically, it triggers strong cell‑cycle arrest, suppresses E2F‑driven transcription, and minimally affects RNA polymerase II phosphorylation, supporting its use in dissecting CDK7‑regulated pathways.
